    Abstract: 盾殼黴菌 (Coniothyrium sp.) 為重要的生物防治菌之一,其二次代謝物具有多種生物活性,如抗病原菌、抑制癌細胞及酵素的產生,因此在醫藥及食品上為新的產品開發菌源。本論文之研究以本土植物棕葉狗尾草 (Setaria palmifolia Stapf ) 莖部分離得到之內生真菌Coni -othyrium sp. CSM進行試驗,經培養基萃取後可得抑制人類肺腺癌 (A549) 及人類肝癌 (Huh 7) 細胞株和病原菌Staphylococcus aureus之產物。再以回應曲面法設計改善培養基組成份,來提升菌絲生長和代謝物產量。固態培養下,培養於溫度30 ℃和培養基起始pH值為4-5時,為此菌之較佳生長條件。經培養基修飾,以添加蛋白?和ZnSO4能有效提升菌絲生長;而添加蛋白?和NaNO3則能提生活性物質產生。另由液態發酵萃取發酵液後可得抑制癌細胞之代謝物質,其萃取物在48小時對肺癌細胞及肝癌細胞生長抑制率IC50為1.12 μg/mL及 0.36 μg/mL。經HPLC純化過程來提升目標代謝物之效益,進一步以發酵槽進行量產測試。目前將以分離純化和結構式分析來得知產物特性,進一步改善培養條件和萃取分離產物方式來提升抑制癌細胞之活性物質產量。
    Some Coniothyrium fungal strains have been reported to have the capability to synthesize secondary metabolites with antibacterial, antifungal, or antialgal activities. An endophytic strain of Coniothyrium sp. was isolated from the stem of palm grass (Setaria palmifolia Stapf) in our laboratory. The extract of this fungal culture exhibited an anti-proliferative effect on cancers and bacteria (Staphylococcus aureus). The objective of this study is to find out the optimal conditions for producing these bioactive compounds. The experimental data have showed that the optimal fermentation temperature was 30℃, and the optimal pH was 4-5. Modified media for the best yield of bioactive cultural extracts were determined, and a set of manipulation parameters for fermentation was decided. The anticancer components of the cultural extracts were partially purified using HPLC technique. The IC50 of the partial purified extracts against human lung adenocarcinoma A549 cells and human hepatocarcinoma Huh 7 cells were 1.12 μg/ml and 0.36 μg/ml after 48 h incubation, respectively. The major anticancer compound of the culture extract has been tried to identify.
