國人的飲食習慣,隨著經濟的成長以及飲食生活的改變,罹患肥胖、高脂血症、心臟病、過敏性氣喘、肝炎及大腸癌等人口急遽上升。本研究鎖定在種子與果實類食材之加工利用,我們已經執行的第一年顯示,將傳統種子類藥材以充氮膨發暨超音波萃取的技術進行加工,對功效分劃之產量有顯著的提高,提高抗過敏免疫調節生物活性,今年度已初步優化各成份分層,計畫中我們已經將多醣分劃經凝膠色層分析分離不同的波峰後分析,在樹突細胞對塵?過敏原調節中,可顯著降低IL-4, IL-13 之分泌,並提高IL-12 的分泌,本製程已初步與專利事務所討論申請製程專利,對未來技轉有信心。今年度的計畫將延用充氮膨發暨超音波萃取的技術再利用酒精為萃取溶媒,則應用於護肝之保健功效,針抗氧化活性高之分劃進行細胞試驗分析,選取最佳分化三組進行動物試驗,委託進行GC-MS-MS 分析並找到特定之結構,希望由此充氮膨發種子暨超音波低溫萃取的方法對於開發新穎功效成份與生物活性有新的發展。 People suffered from obesity, hyperlipidemia, heart disease, allergies, asthma, hepatitis, and rapid population increase in colon cancer were change in eating habits and economic growth in diet. In our previously study, seed and fruit category used nitrogen filled puffing and sonicated extraction techniques for processing, the seeds extracted food ingredients to be separated. Increase or optimize the composition layer to improve the release of water-soluble polysaccharide components and used to modulate allergic effect. In the pre-treat with various separation of polysaccharide in dendritic cells and further stimulated by house dust mite. Dendritic cells demonstrated increased with IL-12, and down-regulated IL-4 and IL-13. In this year, alcohol fraction is applied to study the hepatoprotection. We will use various organic solvents to partition the alcohol fraction and used BNL cell to identify the anti-oxidation potential. We will use three higher anti-oxidation ingredients to conform the hepatoprotective potential by induced by thioacetamide in murine model. We will use GC-MS-MS to identify the structure in active ingredients. We hope that nitrogen filled puffing and low-temperature sonicated extraction will be the efficiency in the development of novel composition and biological activity.
