    Title: 果實種子類中藥抗氧化成分之研究

    Abstract: 本研究針對 100 種果實種子類中藥的抗氧化性進行篩選, 並從具抗氧化 性之中藥分離得到抗氧化成分, 結果摘要如下: (1) 果實種子類中藥之生 育醇含量及抗氧化性首先利用高效液相層析法分析其中的 α-, β-, γ-, 及 δ-生育育醇之含量, 結果發現芡實(Euryale ferox Salisb.)有 最高含量( α- 428.00, β-189.40, γ-38.70, δ-54.50 及總生育醇 710.60μg/g),依此, 選定具抗氧化性且含高量生育醇的芡實, 進行分離 純化, 以了解芡實之抗氧化性與其所含高量生育醇的關聯; 另一方面, 僅 含微量生育醇的補骨脂有很強的抗氧化性, 顯示在補骨脂中尚含其它抗氧 化成分, 針對此點, 亦選定補骨脂做為非生育醇型抗氧化成分之分離對象 。 (2) 芡實之抗氧化成分芡實經正已烷萃取及矽膠管柱層析後, 僅發現 正已烷:乙酸乙酯(95 :5 )沖提部分具抗氧化性, 此部分重複利用高效液 相層析法進行分離純化, 共純化出四種抗氧化成分, 經 UV 及 MS 等分析 法進行構造解析,鑑定其分別為α-, β-, γ- 及 δ-生育醇, 由於其它 部分未顯現強抗氧化性, 因此斷定含量甚豐的生育醇即芡實之主要抗氧化 成分; 同時,在芡實抗氧化成分分離過程中,亦確立一套大量製備各型生育 醇之方法。 (3) 補骨脂之抗氧化成分補骨脂正已烷萃取物經二次矽膠管 柱層析, 具抗氧化性的沖提部分重複以高效液相層析法純化, 得到三種純 化物質, 經紫外線吸光光譜,質譜, 核磁共振光譜等儀器分析法解析, 鑑 定其構造為bakuchiol、an- gelicin 及 psoralen, 再利用硫氰酸鐵法檢 測其抗氧化性, 結果如下:對照組< psoralen(0.2mg)< angelicin(0.2 mg) < α-生育醇(0.2mg) < bakuchiol(0.1mg)< bakuchiol(0.2mg)< bakuchiol(0.3mg)=BHA(0.2mg )< bakuchiol(0.4mg), 由以上結果顯示, bakuchiol之抗氧化性較 α-生育醇強上許多, 甚至已與 BHA 相近。另一 方面, 利用重量法檢測將 bakuchiol 實際應用於 8 種市售食用油中的抗 氧化能力, 結果顯示在各種食用油中均較α-生育醇為好,但此抗氧化性 是來自 bakuchiol 本身的作用, 或來自 bakuchiol 與食用油中所添加抗 氧化劑產生的協同效應, 則仍有待進一步探討。
    The object of this study were to investigate the antioxi- dative activity of 100 Chinese crude drugs of fruits or seeds and to isolate the antioxidative components from some of them. The results were showed as follows. (1) Tocopherol contents and antioxidative activity of 100 Chinese crude drugs from fruits or seeds The results showed that the seeds of Euryale ferox Salisb. had the highest contents ( totol tocopherols 710.60μg/g)among these drugs. The isolation and identification of antioxidative components were undertaken from two antioxidative drugs,E. fe- rox Salisb. having the highest amount of tocopherols, and Pso- ralea corylifolia L. having almost no tocopherol. (2) The isolation and identification of antioxidative compo- nents from Euryale ferox Salisb. The n- hexane extract was fractionated and isolated by vari ous chromatographies. to obtain four antioxidative components. These four isolated components were identified as α-, β-, γ- and δ-tocopherol. (3) The isolation and identification of antioxidative compo- nents from Psoralea corylifolia L. The n- hexane extract was fractionated and isolated by vari ous chromatographies.to obtain three antioxidative components. These three isolated components were identified as bakuchiol, angelicin and psoralen. The antioxidative efficiency of bakuchiol、angelicin and psoralen is in the order of control< Psoralen(0.2mg)<angelicin (0.2mg)< α-tocopherol(0.2mg)< bakuchiol(0.1mg)< bakuchiol(0.2 mg)< bakuchiol(0.3mg)=BHA< bakuchiol(0.4mg). The antioxidative activity of bakuchiol on 8 kinds of the edible oils was more stronger than α-tocopherol.
