    Title: 雷丸抗氧化、螢光成分及揮發性成分之研究

    Abstract: 摘 要 本研究探討雷丸抗氧化及螢光成分,以及雷丸之揮發性成分,主要分成以下二個部分: 1.雷丸抗氧化及螢光成分之萃取、分離及純化 雷丸經磨碎後,所得的粗粉末依序以正己烷、乙酸乙酯及甲醇重覆萃取三次,經過濾收集其濾液,利用旋轉式減壓濃縮機在低於40℃下濃縮,再用硫氰酸鐵法檢測其抗氧化性,結果以甲醇萃取物之抗氧化性較佳,且含螢光成分,因此選取甲醇萃取物來進行第一次XAD-7液相管柱層析分離,而區分為8個區分,將這8個區以分析型高效液相層析找尋良好的分離條件初步分析,將具相似層析圖混合、濃縮,分為三個區分,各區分再經第二次液相管柱層析分離,各沖提區分別用高效液相層析分析,找尋最佳分離條件,重複以高效液相層析純化, 藉各種光譜分析法鑑定,得到四種純化物質,其中三種為7-4’-dihydroxy flavanone-7-O-β-D-glucopyranoside、 4-hydroxymethyl-benzaldehyde及4-hydroxybenzaldehyde另一種推測為1-[2-(linoleoxyl)-4,6-di-hydroxyphenyl]-3-(4-hydroxyphenyl)-1- propanone。其中4-hydroxymethyl-benzaldehyde及1-[2-(linoleoxyl)- 4,6-di-hydroxyphenyl]-3-(4-hydroxyphenyl)-1- propanone在UV燈照射下呈現螢光。 2.雷丸之揮發性成分 雷丸粉末利用水蒸氣蒸餾-溶劑萃取法萃取揮發性成分,並分析其化學組成及檢測揮發性成分與雷丸各層萃取物之抗氧化性;結果顯示,雷丸揮發性萃取物經GC及GC-MS分離鑑定,共鑑定出35種化合物,其中酮類2種、醛類3種、酸類5種、酚類1種、烯類4種、?烯類4種、醇類11種、烷類1種、脂類1種及未知3種;甲醇各種萃取物抗氧化性利用硫氰酸鐵法檢測,所得到的抗氧化性結果為:BHA>甲醇萃取物>α-生育醇>水蒸氣蒸餾-溶劑萃取法>乙酸乙酯萃取物>正己烷萃取物>對照組。
    ABSTRACT The purposes of this were to investigate antioxidative, fluorescent and volatile components from Nucleoid of Polyporus mylittae. The results were summarized as following: (1) The antioxidative components from the methanol extract of Nucleoid of Polyporus mylittae The ground powder of Nucleoid of Polyporus mylittae was extracted with n-hexane, ethyl acetate and methanol. The antioxidative activity of the extracts were measured by the ferric thiocyanate method. The antioxidative efficiency was found in the order: BHA > methanol extract > α-tocopherol > ethyl acetate extract > n-hexane > control. The methanol extract had strong antioxidative activity and contained fluorescent components. The methanol extract was fractionated and isolated by various chromatographies to obtain four pure compounds. Three of the isolated components were identified to be 7-4’-dihydroxy flavanone-7-O-β-D-glucopyranoside, 4-hydroxymethyl-benzaldehyde and 4-hydroxybenzaldehyde. The other one was tentatively concluded to be 1-[2-(linoleoxyl)-4,6-di-hydroxyphenyl]-3- (4-hydroxyphenyl)-1- propanone. Two of the four pure compounds which were 4-hydroxymethyl- benzaldehyde and 1-[2-(linoleoxyl)- 4,6-di- hydroxyphenyl] -3-(4-hydroxyphenyl)-1- propanone had fluorescence. (2) Volatile components from Nucleoid of Polyporus mylittae The volatile extracts were obtained from Nucleoid of Polyporus mylittae by Likens-Nickerson extraction. The volatile extract was isolated and identified by gas chromatography (GC) and gas chromatography mass spectrometry (GC-MS). The 28 components of the volatile extracts had been identified, including 11 alcohols, 5 acids, 4 alkylenes, 4terpene, 3 aldehydes, 2 ketones, 1 phenol, 1 alkane, 1 esters and 3 unknow. The antioxidative activities of the volatile extracts and solvents extracts were analyzed by the ferric thiocyanate method and compared with BHA andα-tocopherol. The relative activity was BHA > methanol extract > α-tocopherol > volatile extracts >ethyl acetate extract > n-hexane > control.
