    Abstract: 本研究主要目的在探討靈芝液態培養時,各種不同物理及化學因子對 靈芝菌絲體生長與多醣生成的影響. 主要實驗於250ml三角瓶,500ml三 角瓶及攪拌式生化反應器中進行.以不同植物性原料為氮源實驗中,花生粉 所得到之菌體及多醣量最高.以添加固形物酒糟粕做為氮源培養時,所得到 之菌絲濃度與多醣濃度均比添加無機氮源氯化銨高.於接菌量方面,接菌量 愈高則菌體產量愈高,但多醣量無明顯增加.於添加油類實驗中,顯示添 加0.3%橄欖油可使菌體產量提昇,而添加0.15%紅花籽油則可刺激多醣生 成,但這兩種油類添加量超過1%,則會造成抑制作用.利用酒糟水當成氮源 培養時,發現酒糟水濃度為20%對於菌體量或多醣量均有良好效果,但若酒 糟水培養基中最初所含的總酸與總胺基酸量太高,將不利於菌絲與多醣生 成.於攪拌式生化反應器培養方面,在溫度為30度,200rpm&1vvm之條件下為 一理想操作環境,培養八天時多醣濃度可達52.8mg/100ml. 本研究結果 証實以三角瓶與攪拌式生化反應器進行液態培養,可以有效縮短靈芝培養 時間,而且能夠快速生成菌絲體與含多醣之發酵液.
    The main purpose of this research was to investigate the influence ofdifferent environmental factors on the production of mycelium and poly-saccharide in submerged culture of Ganoderma lucidum.The culture tests were carried out in shake flasks and stirred-tank fermentor. The results indicated that peanut powder was the best N-source forthe production of mycelia or polysaccharide.Distiller'' grain was betterthan ammonium chloride.Higher inoculum ratio was believed to be advanta-geous for mycelial growth rate,but not for the formation of polysaccharide.The addition of natural oils into the medium was demonstrated to have greatinfluence on the formation of mycelia or polysaccharide.However,excessamount could cause inhibition effect.Similar results was found when thin stillage was used as N-source.The optimal operating conditions for theculture of G. lucidum in stirred-tank fermentor were determined to bearound 200rpm&1vvm, and the concentration of polysaccharide reach to52.8mg/100ml at 8th day. The results from this research demonstrate that compared with solidculture,submerged culture of G.lucidum in flasks or stirred-tank fermentorcould shorten the period of mycelial growth & polysaccharide formation.
