    Abstract: 本研究主要目的,為探討以三角瓶液態培養,了解不同添加物及培養條件對於樟芝(Antrodia camphorata BCRC35396 )菌絲生長、抗氧化效果、DPPH自由基掃除活性及總多酚含量之影響。實驗結果顯示:於250ml三角瓶中,以100ml培養液含玉米澱粉4.78%、YM Broth 3.19%在pH=5.54、25℃下,以轉速100rpm於恆溫培養箱培養14天,其中添加10%之乳化劑(tween85)菌體乾重達2.39g/100ml,是未添加的2倍左右,添加0.1% polypropylene glycol掃除DPPH自由基活性達39.93%,較未添加提高10%;添加5%葡萄籽油總多酚量可達27.9mg/g extract,是未添加添加的1.5倍,0.1%polypropylene glycol總多酚量可達68.9mg/g extract,是未添加的3倍。而碳氮比愈低,菌絲生長情況愈好,在碳源濃度5%、碳氮比0.78:1下,菌絲乾重最高可達1.52g/100ml,是碳氮比50:1的2.5倍,且總多酚含量與掃除DPPH自由基活性有相關性。
    The main purpose of this research was to study the effect of culture conditions on the mycelium production, the formation of antioxidant activity in submerged culture of Antrodia camphorata (BCRC35396).Shaken-flask cultures were carried out in the medium containing 4.78% of corn starch, 3.19% of YM broth at 25℃, initial pH 5.54 and 100rpm. The supplementation of 10% tween 85 was found to be beneficial to the mycelium growth. The biomass concentration reached 2.39g/100ml, which was two folds higher than that of the control. With the addition of 0.1% polypropylene glycol, DPPH radical scavenging activity could rise to 39.93% and the amount of total polyphenol could reach 68.9(mg/g extract), which was 3 times higher then that of the control. Moreover, 5% of grape seed oil could cause a 1.5 time increase of the amount of total polyphenol.The media with low C/N ratio was proved to be favorable to the mycelium growth and the biomass concentration could reach 1.52g/100ml at low C/N ratio of 0.78. Compared with shaken-flask cultures, surface cultures could lead to the increase of DPPH radical scavenging activity and antioxidant activity.
